Physically, maybe. The road is used for lumber trails...only open during the dry season part of the year. You would not want to try this though, due to the heavy bombardment of Colombian and Panamanian rebels/guerillas. There is no police enforcement in the thickness of the Darien rainforest.

I doubt you'd be able to go thru the gap. However, a Ferry ride to the city of Turbo in Colombia might be possible. Or, you could try to drive along the Atlantic, along the coast of the Golfo de Uraba.
In any case, you would need to get to the city of Turbo to catch a main road heading into the interior of Colombia.

I understand there aren't any main roads connecting the interior of Panama with Colombia, but aren't there at least dirt roads?

yes, it is posible but due to certain activity, not a good idea.
so, you could also consider driving to a certain point in panama and taking a ferry to colombia with your car.
